Fig. 6 Peritoneal macrophages were harvested from mice (9-14 week-old, female) 4 days after intraperitoneal injection of 2 ml of 10 % proteose peptone solution (Difco, Detroit). Cells were cultured in RPMI 1640 medium supplemented with 2 % fetal calf serum. The medium was aspirated and replaced with medium containing the Ca2+ ionophore A23187 (5 micro molar) or lipopolysaccharide (LPS, 1 micro g/ml). Cells were stimulated with A23187 for 10 min or with LPS for 12 hours for PGE2 and Cys-LT measurements or 30 min for PAF measurement. The medium was collected and PGE2 and cysteinyl-leukotrienes (Cys-LT) were quantified using enzyme immunoassay (EIA) kits (Amersham). Levels of PAF in the samples containing the medium and cells were measured using a radioreceptor binding assay. Amounts of eicosanoids and PAF in the absence of stimulations are labeled "control".
